c-Fos over-expression promotes radioresistance and predicts poor prognosis in malignant glioma

چکیده
c-Fos is a major component of activator protein (AP)-1 complex. It has been implicated in cell differentiation, proliferation, angiogenesis, invasion, and metastasis. To investigate the role of c-Fos in glioma radiosensitivity and to understand the underlying molecular mechanisms, we downregulated c-Fos gene expression by lentivirus-mediated shRNA in glioma cell lines and subsequently analyzed the radiosensitivity, DNA damage repair capacity, and cell cycle distribution. Finally, we explored its prognostic value in 41 malignant glioma patients by immunohistochemistry. Our results showed that silencing c-Fos sensitized glioma cells to radiation by increasing radiation-induced DNA double strand breaks (DSBs), disturbing the DNA damage repair process, promoting G2/M cell cycle arrest, and enhancing apoptosis. c-Fos protein overexpression correlated with poor prognosis in malignant glioma patients treated with standard therapy. Our findings provide new insights into the mechanism of radioresistance in malignant glioma and identify c-Fos as a potentially novel therapeutic target for malignant glioma patients.

Dysregulation of Fra1 expression by Wnt/β-catenin signalling promotes glioma aggressiveness through epithelial–mesenchymal transition
Aberrant expression of Fos-related antigen-1 (Fra1) is commonly elevated in various malignant cancers and is strongly implicated in invasion and metastasis. However, the molecular mechanisms underlying its dysregulation in human glioma remain poorly understood. In the present study, we demonstrate that up-regulation of Fra1 plays a crucial role in the glioma aggressiveness and epithelial-mesenc...
متن کاملCofilin-1 and phosphoglycerate kinase 1 as promising indicators for glioma radiosensibility and prognosis
Glioma is a primary malignancy in central nervous system. Radiotherapy has been used as one of the standard treatments for glioma for decades. Since radioresistance can reduce the curative efficacy of radiotherapy in glioma, investigating the cause of radioresistance and predicting the tumour radiosensibility appeared particularly important. We previously reported that CFL1 and PGK1 are over-ex...

Background: Hepatocellular carcinoma is the most common primary malignant tumor of the liver. The effect of some genes especially those involved in cell cycle regulation have been shown in the development of this cancer in several studies but there are some controversies about them yet. Materials and methods: The paraffin-embedded tissue samples of 25 patients (18 males and 7 females) with hep...
متن کاملO-35: Over-Expression of XRCC1 As Potential Biomarker for Poor Prognosis in Human Preimplantation Embryos: Selection by Study of 84 Genes Involved in DNA Damage Signaling Pathways
Background: Chromosome abnormalities are associated with poor morphology and development in human preimplantation embryos, all together lead to poor outcomes. This study aimed to explore altered expression of DNA damage pathways in “poor morphological and development embryos with sever aneuploidies”. Materials and Methods: Surplus day-4 embryos of PGD cases were pooled in two groups: Poor progn...
متن کاملCAVEOLIN-1 expression in brain metastasis from lung cancer predicts worse outcome and radioresistance, irrespective of tumor histotype
Brain metastases develop in one-third of patients with non-small-cell lung cancer and are associated with a dismal prognosis, irrespective of surgery or chemo-radiotherapy. Pathological markers for predicting outcomes after surgical resection and radiotherapy responsiveness are still lacking. Caveolin 1 has been associated with chemo- and radioresistance in various tumors, including non-small-c...
